Proteogenic and non-proteogenic alpha amino acids are commonly analyzed and quantified for various application. The MetaSci Amino Acids Standard Mixture Kit is designed to accelerate the identification and quantification by packing as many as compatible compounds in a mix, allowing multiple spectra recording in a single injection. The kit is composed of 60 amino acids, in three mixes. Each mix has 20 amino acids in it.  An internal standard, 

2-Ethoxyisonicotinic acid, is used in all mixes at the constant concentration of 50 ug/mL to normalize the variance in volume of injection. This internal standard is highly ionizable both at positive and negative mode, resulting in consistent response. 

 

The kit comes in two different types. The first type contains only the mixtures with each amino acid at 100ug/mL. The second type comes in a serially diluted series of mixtures starting at 100ug/mL followed by 50, 25, 10, 5, 2.5, 1.0, 0.5, 0.25 and 0.10 ug/mL solutions. The advantage of using the serial dilution is two folds: a) higher accuracy because the dilutions are made in controlled environment, with calibrated equipment and in higher volumes, resulting in lower inaccuracy in preparation;  b) all the dilutions are made with a buffer containing 50ug/mL of IS. The constant presence of IS at certain concentration from stock solutions to the last dilution makes it possible to accurately calculate any variance in the system and normalize the results. 

Another feature of this kit is the absence of any isobaric interference. Every amino acid molecule in each mix is at least 1 Da different from all other molecules which eliminates uncertainties in identification of molecules after changing method, column, solvent, pH, etc. For example, the well-known isobaric Leu and Ile are present in two different mixes, which removed the guesswork. Cystein and Homocystein are included in the kit and their accurate amount can be measured by calcuating the concentration of their dimers Cystine and Homocystine which is provided in calibrant series in a third ampule.  

 

A kit comes in 3 mixture solutions. Each mix comes in 5 microampule of 0.2 uL quantity (total of 1.0 mL). The calibrant series come in 5 microampules of 0.2 uL for each calibration point. There are a total of 10 calibration point (100ug/mL - 0.1ug/mL).

 

We provide multiple methods of identification on HILIC in our interactive online Certificate of Analysis. The list of compounds with annotations and exact masses can be downloaded as a .csv file for fast transfer of information to your software. The top two transitions are also provided for all the amino acids in the CoA.

 

All of the CoA, SDS and methods come in digital format at our specification page which is product specific. No printed material is provided with this kit. The hard copy SDS is available upon request.  

 

You will need 9-425 vials (common autosampler vials)  and caps to be able to run the samples directly. If your autosampler does not accept this type of vials, you will need to pipette out the solution to the compatible vessel. If you use plate based auto sampler, we can provide a custom 96 well plate that fits the micro ampoule for direct injection.
